hwp1 gene expression of candida albicans and study its role in adherence

چکیده
candida albicans is an opportunistic fungal pathogen found as mycoflora in the human body surfaces. sevral genes play a crucial rule in its virulence including hwp1 (hyphal wall protein 1), bcr1 and als gene family. hwp1 gene is a responsible for coding a cell surface protein, which mediates biofilm formation in candida albicans. here we investigated the presence of the hwp1gene was characterised among candida albicans isolates in women with recurrent vaginal canididasis and further we studies its role in cell adherence. we used 50 candida albicans clinical isolates resistant to fluconazole. rna (of samples were extracted using glass bead and lysis buffer and cdna was synthesized using reverse transcriptase enzyme. expression of (hwp1) gene was analysed using reverse transcriptase-plolymerase chain reaction (rt-pcr). the ability of adherence of the isolates with or without the expression of hwp1 were characterized using hela cells. statistical analysis were performed using t-test and two-way anova spss software. our results showed that the hwp1 gene were present in 47 samples (94%) out of 50 isolates, 27 samples (57%) had expression of hwp1. the result of adherence assay revealed that the isolates with the expression of hwp1 gene and control isolates was statistically different (p

متن کاملInhibitory Effect of Carvacrol on the Expression of Candida albicans Hyphae-Specific Gene (HWP1)
BACKGROUND AND OBJECTIVE: Candida albicans infection is a problem of growing clinical importance, particularly in immunocompromised populations. This study aimed to investigate the inhibitory effect of carvacrol on the hyphae formation and expression of C. albicans HWP1. METHODS: This cross-sectional study was done over a 6-month period in 2016-2017. Vaginal, mouth and skin surface swabs were...
متن کاملRole of actin cytoskeletal dynamics in activation of the cyclic AMP pathway and HWP1 gene expression in Candida albicans.
Changes in gene expression during reversible bud-hypha transitions of the opportunistic fungal pathogen Candida albicans permit adaptation to environmental conditions that are critical for proliferation in host tissues. Our previous work has shown that the hypha-specific adhesin gene HWP1 is up-regulated by the cyclic AMP (cAMP) signaling pathway. However, little is known about the potential in...
متن کاملCandida albicans HWP1 gene expression and host antibody responses in colonization and disease.
In vivo expression of the developmentally regulated Candida albicans hyphal wall protein 1 (HWP1) gene was analysed in human subjects who were culture positive for C. albicans and had oral symptoms (n=40) or were asymptomatic (n=29), or had vaginal symptoms (n=40) or were asymptomatic (n=29). HWP1 mRNA was present regardless of symptoms, implicating hyphal and possibly pseudohyphal forms in muc...
متن کاملQuantitation of ergosterol content and gene expression profile of ERG11 gene in fluconazole-resistant Candida albicans
Background and Purpose: The frequency of opportunistic fungal infections in immunocompromised patients, especially by Candida species, has sharply increased in the last few decades. The objective of this study was to analyse the ergosterol content and gene expression profiling of clinical isolates of fluconazole resistant Candida albicans. Materials and Methods: Sixty clinical samples were ide...
متن کاملEffect of 900 MHz microwave radiation on alpha-int1 gene expression, proliferation and adherence of Candida Albicans
To date, registered users of mobile phone communication network exceeded from total numbers of the world population, while a little knowledge of the biological effects of, 900-1800 MHz microwave radiation, originating from the handsets or the base transceiver stations, have been released. The current study was designed for evaluation of 900-MHz radiation effects on Candida albicans proliferatio...
